Product Description

In order to confirm cytotoxicity, evaluation using only live cells or only dead cells as indicators may not be sufficiently reliable, and therefore, there are an increasing number of cases where experiments are supported by evaluating multiple indicators based on different measurement principles.

This kit is a set of two different indicators, Cell Counting Kit-8 (CCK-8) and Cytotoxicity LDH Assay Kit-WST (LDH Assay Kit). An assay manual is available to evaluate these two assays using the same sample.

Technical info

When confirming cytotoxicity, evaluation using only live cells or only dead cells as indicators may not be sufficiently reliable, and therefore, more and more experiments are being supported by evaluation using multiple indicators based on different measurement principles.

Experimental Example

Cell Counting Kit-8 and Cytotoxicity LDH Assay Kit-WST measure dehydrogenase activity and released amount of LDH as reflective of living cells and dead cells, respectively. According to the results with anticancer drug, the number of living cells was reduced, and the number of dead cells increased as the concentration of Mitomycin C increased. Interestingly, a low concentration of Mitomycin C (green frame) inhibits cell proliferation earlier than inducing cell death unlike a high concentration (red frame).

Measuring Principle

LDH Assay Kit: The activity of LDH leaked out of cells due to cell membrane damage can be confirmed as an indicator of dead cells.
Cell Counting Kit-8: Metabolic activity in living cells can be confirmed using NADH as an indicator.

The same cell sample can be applied

This kit includes CCK-8 for measuring live cells and LDH Assay Kit for measuring dead cells, both of which can be evaluated by plate assay at the same absorbance wavelength. The same cell sample can be used to evaluate both live and dead cells at the same time, saving time and labor.
